1945년 아일랜드 웩스퍼드에서 태어났다. 열두 살 때 제임스 조이스의 『더블린 사람들』을 읽고 영향받아 처음으로 소설을 쓰기 시작했고, 미술과 건축에 관심을 쏟았다. 세인트 피터스 칼리지를 졸업한 뒤 아일랜드 항공에 취직했고, 1969년 <아이리시 프레스>에 입사해 <아이리시 타임스>로 이직, 1999년까지 기자생활과 작품활동을 병행했다. 1970년 작품집 『롱 랭킨』을 발표하며 작가로서 첫발을 내디뎠다. 이후 발표한 두 편의 장편소설에 ‘아일랜드 소설’이라는 평가가 따르자 새로운 작풍과 주제에 몰두하며 ‘과학 4부작’ 『닥터 코페르니쿠스』 『케플러』 『뉴턴 레터』 『메피스토』와 ‘예술 3부작’ 『증거의 책』 『유령들』 『아테나』를 잇달아 출간해 평단과 독자의 지지를 얻었다. 2005년 발표한 장편소설 『바다』로 유례없이 경합이 치열했던 그해 맨부커상을 수상하며, 제임스 조이스와 사뮈엘 베케트의 뒤를 잇는 아일랜드 최고의 작가로 자리매김했다. 2006년부터 ‘벤저민 블랙’이라는 필명으로 범죄소설과 대체역사소설을 발표하다가, 2020년 『눈』부터는 모든 소설을 존 밴빌 명의로 출간하고 있다. 2012년 『오래된 빛』으로 ‘앨릭스와 캐스 클리브 3부작’을 마무리하며 다시금 평단의 찬사와 함께 아일랜드 도서상을 받았다. 가디언 소설상, 래넌 문학상, 프란츠 카프카 상, 유럽문학상, 아스투리아스 왕세자상 등을 수상한 밴빌은 매년 노벨문학상 후보로도 거론되고 있다.
