1828년 러시아 야스나야 폴랴나에서 톨스토이 백작 집안의 넷째 아들로 태어났다. 1844년 카잔 대학교에 입학하나 대학 교육에 실망, 1847년 고향으로 돌아갔다. 진보적인 지주로서 새로운 농업 경영과 농노 계몽을 위해 일하려 했으나 실패로 끝나고 이후 삼 년간 방탕한 생활을 했다. 1851년 맏형이 있는 캅카스로 가서 군대에서 복무했다. 이듬해 잡지 《소브레멘니크》에 익명으로 「유년 시절」 연재를 시작하면서 작가로서 첫발을 내디뎠다. 작품 집필과 함께 농업 경영에 힘을 쏟는 한편, 농민의 열악한 교육 상태에 관심을 갖게 되어 학교를 세우고 1861년 교육 잡지 《야스나야 폴랴나》를 간행했다. 1862년 결혼한 후 문학에 전념하여 『전쟁과 평화』, 『안나 카레니나』 등 대작을 집필, 작가로서의 명성을 누렸다. 그러나 이 무렵 삶에 대한 회의에 시달리며 정신적 위기를 겪었다. 그리하여 1880년 이후 원시 기독교 사상에 몰두하면서 사유재산 제도와 러시아 정교에 비판을 가하고 『교의신학 비판』, 『고백록』 등을 통해 ‘톨스토이즘’이라 불리는 자신의 사상을 체계화했다. 또한 술과 담배를 끊고 손수 밭일을 하는 등 금욕적인 생활을 지향하며 빈민 구제 활동을 하기도 했다. 민중이 쉽게 읽을 수 있도록 민담 22편을 썼는데 그중에서도 「인간에게 많은 땅이 필요한가」는 소설가 제임스 조이스가 “문학사에서 가장 위대한 이야기”로 꼽기도 했다. 1899년 종교적인 전향 이후의 대표작 『부활』을 완성했다. 사유재산 및 저작권 포기 문제로 시작된 아내와의 불화 등으로 고민하던 중 1910년 집을 떠나 폐렴을 앓다가 아스타포보 역장의 관사에서 영면하였다.
